质谱流式细胞技术(Mass Cytometry, CyTOF)采用带有金属同位素标签的抗体对细胞进行标记,通过飞行时间质谱分析各细胞上的标签组成,进行细胞表型和功能的深入研究。

CyTOF和流式的区别?
简单来说CyTOF就是高通量流式,采用金属同位素标签替代传统流式的荧光标签来标记细胞,并通过飞行时间质谱来检测每个细胞上金属标签的精确含量。和传统流式相比,CyTOF不会存在“串色”的问题,不需要补偿计算,因此可以同时检测更多种蛋白,普罗亭目前能够同时测42个marker,而流式很难超过20个marker,所以CyTOF可能发现流式难以观察到的一些价值信息。
CyTOF能检测什么样本?
组织和外周血都可以测,也可以直接提供细胞悬液或者细胞系。
CyTOF的样本要求有什么?
有细胞数量的要求,血样的细胞数量要达到3×10^6,而组织,消化后的细胞数量要达到1×10^6。此外,新鲜样本需要在取样后48小时内寄送至普罗亭实验室。
